Ledent<\/strong><\/p>\n<p>Nos recherches sur l&rsquo;association possible entre l&rsquo;exposition \u00e0 des champs magn\u00e9tiques de tr\u00e8s basse fr\u00e9quence et un risque accru de maladie d&rsquo;Alzheimer ont \u00e9t\u00e9 compl\u00e9t\u00e9es par une \u00e9tude cytog\u00e9n\u00e9tique utilisant les m\u00e9thodes d&rsquo;hybridation in situ par fluorescence (FISH). Avec cette \u00e9tude, nous n&rsquo;avons pas pu confirmer les r\u00e9sultats ant\u00e9rieurs et n&rsquo;avons donc pas trouv\u00e9 d&rsquo;arguments en faveur d&rsquo;un risque accru de maladie d&rsquo;Alzheimer li\u00e9 \u00e0 l\u2019exposition aux champs magn\u00e9tiques d\u2019extr\u00eamement basses fr\u00e9quences.<\/p>\n<p>Il n\u2019existe \u00e0 ce jour aucune indication solide d\u2019un effet des champs magn\u00e9tiques de fr\u00e9quences extr\u00eamement basses sur l&rsquo;apparition ou l&rsquo;\u00e9volution de la maladie d&rsquo;Alzheimer.[\/vc_column_text][vc_custom_heading text=\u00a0\u00bb2013-2017&Prime; font_container=\u00a0\u00bbtag:h3|text_align:left|color:%233f3f3f\u00a0\u00bb use_theme_fonts=\u00a0\u00bbyes\u00a0\u00bb][vc_column_text]<strong>L. Anthonissen<\/strong><\/p>\n<p>Nos recherches sur l&rsquo;association possible entre l&rsquo;exposition aux champs magn\u00e9tiques d&rsquo;extr\u00eamement basses fr\u00e9quences (CM-EBF) et un risque accru de maladie d&rsquo;Alzheimer sont bas\u00e9es sur des \u00e9tudes (cyto)g\u00e9n\u00e9tiques de lign\u00e9es cellulaires humaines. Nous avons relev\u00e9 des aberrations chromosomiques \u00e0 des niveaux d&rsquo;exposition sup\u00e9rieurs \u00e0 100\u00b5T. Cependant, nous avons aussi trouv\u00e9 certaines indications de dommages g\u00e9n\u00e9tiques \u00e0 des niveaux d&rsquo;exposition de \u2265 50 \u00b5T, vraisemblablement des perturbations de la division cellulaire et de l&rsquo;amplification g\u00e9n\u00e9tique. Ce r\u00e9sultat est indicatif d&rsquo;un effet qui pourrait correspondre \u00e0 ceux observ\u00e9s chez les patients atteints d&rsquo;Alzheimer. Il est toutefois trop t\u00f4t pour tirer des conclusions et actuellement nous am\u00e9liorons notre recherche en menant des \u00e9tudes approfondies.\u00a0 Les r\u00e9sultats ne sont pas encore disponibles.<\/p>\n<p>Enfin, nous avons \u00e9galement constat\u00e9 des dommages g\u00e9n\u00e9tiques moindres par rapport \u00e0 ceux observ\u00e9s dans les cellules non expos\u00e9es \u00e0 de faibles niveaux d&rsquo;exposition (&lt;10 \u00b5T), ce qui peut indiquer une \u00ab\u00a0r\u00e9ponse adaptative\u00a0\u00bb. On ne sait pas encore si cet effet biologique b\u00e9n\u00e9fique \u00e0 premi\u00e8re vue persiste lorsque les cellules\/organismes sont expos\u00e9s pendant de longues p\u00e9riodes. Moukhliss<\/strong><\/p>\n<p>Nous analysons l\u2019association entre l\u2019exposition aux champs magn\u00e9tiques de fr\u00e9quences extr\u00eamement basses et l\u2019augmentation du risque de maladie d\u2019Alzheimer \u00e0 l\u2019aide d\u2019\u00e9tudes (cyto)g\u00e9n\u00e9tiques sur des lign\u00e9es de cellules humaines. Nous avons port\u00e9 une attention particuli\u00e8re sur de possibles effets g\u00e9n\u00e9tiques coop\u00e9ratifs avec des environnementaux carcinog\u00e8nes\/mutag\u00e8nes, et \u00e0 l\u2019induction d\u2019anomalies similaires aux dommages g\u00e9n\u00e9tiques relev\u00e9s dans les cellules de patients atteints d\u2019Alzheimer. Dans ce but, nous avons valid\u00e9 le test de cytome, utilis\u00e9 le test bact\u00e9rien VITOTOX pour analyser les dommages \u00e0 l\u2019ADN r\u00e9sultant d\u2019effets coop\u00e9ratifs avec des agents mutag\u00e8nes (divers modes d\u2019action) et r\u00e9alis\u00e9 le test de cytome sur des cellules humaines C3A et SH-SY5Y expos\u00e9es \u00e0 des champs magn\u00e9tiques EBF. Nous travaillerons \u00e9galement sur des cellules IMR-32, mais les analyses doivent encore \u00eatre finalis\u00e9es.<\/p>\n<p>A ce jour, nos r\u00e9sultats sont les suivants :<\/p>\n<ul>\n<li>Aucun signe d&rsquo;effet coop\u00e9ratif (champ magn\u00e9tique 50 Hz +\u00a0agents chimiques carcinog\u00e8nes ou mutag\u00e8nes) n&rsquo; a \u00e9t\u00e9 trouv\u00e9 avec le test VITOTOX. Cela est quelque peu en contradiction avec nos r\u00e9sultats ant\u00e9rieurs.\u00a0En 2003, ce test avait permis de montr\u00e9 des effets d\u2019une co-exposition avec une substance aneug\u00e9nique sur des globules blancs, mais les effets \u00e9taient limit\u00e9s.<\/li>\n<li>Nous avons relev\u00e9 une augmentation du nombre de cellules pr\u00e9sentant de grands micronoyaux (indicateurs d\u2019une action aneug\u00e9nique) et des bourgeons nucl\u00e9aires (indicateurs d\u2019une amplification g\u00e9nique). Comme attendu, aucun aberrations de la structure des chromosomes n\u2019a \u00e9t\u00e9 observ\u00e9e (ponts nucl\u00e9aires, petits micronoyaux).<\/li>\n<li>Pas d\u2019effet clair sur la prolif\u00e9ration cellulaire (index de division cellulaire) et sur l\u2019apoptose (mort cellulaire programm\u00e9e). A 500 \u00b5T, la prolif\u00e9ration des cellules \u00e9tait l\u00e9g\u00e8rement sup\u00e9rieure (index plus \u00e9lev\u00e9 de division cellulaire), mais sans atteindre le seuil de signification statistique. Dans un travail ant\u00e9rieur sur des lymphocytes (1), nous avions observ\u00e9 des effets sur la prolif\u00e9ration cellulaire \u00e0 de hauts niveaux d\u2019exposition (800\u00b5T).<\/li>\n<li>Aucune r\u00e9ponse cellulaire sp\u00e9cifique n\u2019a \u00e9t\u00e9 observ\u00e9e dans le sens o\u00f9 les cellules C3A et SH-SY5Y donnent apparemment la m\u00eame r\u00e9ponse lors de l\u2019exposition aux champs magn\u00e9tiques d\u2019extr\u00eamement basses fr\u00e9quences. Toutefois, les lymphocytes humains semblent avoir une r\u00e9ponse diff\u00e9rente (en accord avec nos r\u00e9sultats de 2003).<\/li>\n<li>Gr\u00e2ce \u00e0 la technique de coloration FISH (Fluorescence in situ hybridization), nous pourrons confirmer (ou non) l\u2019effet sur l\u2019aneuplo\u00efdie (perturbations de la division cellulaire) et l\u2019\u00e9ventuelle implication des chromosomes 21 et 17 sur lesquels sont situ\u00e9s les g\u00e8nes amylo\u00efde \u03b2 et Tau. Ces deux g\u00e8nes sont consid\u00e9r\u00e9s comme tr\u00e8s importants dans le processus de d\u00e9veloppement de la maladie d\u2019Alzheimer. Ces analyses, ainsi que la finalisation des exp\u00e9riences en cours, sont nos priorit\u00e9s des 9 prochains mois.<\/li>\n<li>Les courbes \u201cdose-r\u00e9ponse\u201d montrent une trajectoire non monotones. Bien qu\u2019un grand nombre de substances semblent \u00eatre capables de produire un tel effet (par exemple, les hormones naturelles, les polluants environnementaux, les plastiques, les phytooestrog\u00e8nes et les pesticides), nous ne savons pas comment expliquer ce r\u00e9sultat en ce qui concerne les CM-EBF.<\/li>\n<\/ul>\n<p>Il convient de souligner que malgr\u00e9 certains r\u00e9sultats, voir plus haut, il n\u2019existe \u00e0 ce jour aucune indication solide d\u2019un effet des champs magn\u00e9tiques de fr\u00e9quences extr\u00eamement basses sur l&rsquo;apparition ou l&rsquo;\u00e9volution de la maladie d&rsquo;Alzheimer.<\/p>\n<p>(1) Verheyen G., Pauwels G., Verschaeve L., &amp; Schoeters G.
