P.Mineur<\/strong><\/p>\n<p>Afin de d\u00e9terminer si l&rsquo;exposition \u00e0 des champs \u00e9lectromagn\u00e9tiques (CEM) pourrait entra\u00eener des cons\u00e9quences biologiques et sanitaires, il s&rsquo;agit de <strong>comprendre le ou les m\u00e9canismes potentiels d&rsquo;interaction avec les structures biologiques<\/strong>. Un m\u00e9canisme plausible serait l&rsquo;interf\u00e9rence des CEM avec les \u00e9v\u00e9nements qui se produisent au niveau de la membrane cellulaire et qui permettent la communication des cellules avec l&rsquo;environnement ext\u00e9rieur (hormones, facteurs de croissance&#8230;) \u00e0 travers des r\u00e9cepteurs (antenne). La transmission de tels messages est r\u00e9alis\u00e9e par des seconds messagers (les ions calcium, l&rsquo;AMP cyclique&#8230;) pour, en finalit\u00e9, r\u00e9guler les fonctions cellulaires (prolif\u00e9ration, fabrication des produits d&rsquo;exportation, migration, mort cellulaire programm\u00e9e&#8230;). Des perturbations dans cette cascade d&rsquo;\u00e9v\u00e9nements peuvent avoir des cons\u00e9quences sur la sant\u00e9.<\/p>\n<p>Des recherches in vitro, avec l&rsquo;utilisation d&rsquo;un grand nombre de type de cellules dans des conditions contr\u00f4l\u00e9es, ont \u00e9t\u00e9 r\u00e9alis\u00e9es de part le monde afin d&rsquo;analyser les effets des CEM sur ce r\u00e9seau de communication avec des r\u00e9sultats n\u00e9gatifs et positifs, qui n&rsquo;ont pas pu, dans certain cas, \u00eatre reproduits par d&rsquo;autres \u00e9quipes. Notre \u00e9quipe a observ\u00e9 une modulation par les CEM des signaux calciques induite par des facteurs de croissance dans des cellules humaines normales. Cet effet a uniquement \u00e9t\u00e9 observ\u00e9 \u00e0 certains niveaux de concentration des facteurs de croissance dans le s\u00e9rum qui activaient de mani\u00e8re sous-optimale les cellules. Un autre m\u00e9canisme potentiel sugg\u00e9rerait que les CEM, non mutag\u00e9niques par eux-m\u00eame, pourraient agir en tant que co-carcinog\u00e8ne. Notre approche a \u00e9t\u00e9 de combiner les CEM \u00e0 un facteur environnemental reconnu comme nuisible, les radiations UV, responsables de l&rsquo;augmentation de l&rsquo;incidence de cancers agressifs au niveau de la peau tel que le m\u00e9lanome. Jusqu&rsquo;\u00e0 pr\u00e9sent, nous n&rsquo;avons <strong>pas mis en \u00e9vidence d&rsquo;alt\u00e9ration de la r\u00e9ponse cellulaire aux UV en pr\u00e9sence des CEM<\/strong>.<\/p>\n<p>Le seul effet des CEM que nous avons pu reproduire concerne la conservation des oscillations de la concentration du Ca2+ dans des cellules activ\u00e9es par des concentrations sous-optimales de facteurs de croissance s\u00e9riques. L&rsquo;implication de mol\u00e9cules du signal calcique de la famille des MAPKinases, ERK \u00bd, doit \u00eatre confirm\u00e9e. Les autres membres de cette famille, p-38 and JUN-kinase, doivent \u00eatre analys\u00e9s. Les recherches utilisant siRNA contre MAPKinases devront confirmer leur r\u00f4le dans le signal calcique et les effets des CEM. La signification de cette observation sur la prolif\u00e9ration cellulaire (survie) et la mort cellulaire programm\u00e9e (apoptose) sera \u00e9valu\u00e9e sur d&rsquo;autres types de cellules, normales et transform\u00e9es.<\/p>\n<p>Jusqu&rsquo;\u00e0 pr\u00e9sent, aucune influence significative des CEM sur l&rsquo;expression des g\u00e8nes induits par les UVB, potentiellement impliqu\u00e9s dans l&rsquo;angiogen\u00e8se, n&rsquo;a \u00e9t\u00e9 observ\u00e9e, ni sur des fibroblastes provenant de derme humain, ni sur des k\u00e9ratinocytes provenant de l&rsquo;\u00e9piderme humain. La r\u00e9p\u00e9tition de ces exp\u00e9rimentations sur des k\u00e9ratinocytes poss\u00e9dant l&rsquo;oncog\u00e8ne ras sera envisag\u00e9e.[\/vc_column_text][vc_custom_heading text=\u00a0\u00bb1999-2000&Prime; font_container=\u00a0\u00bbtag:h3|text_align:left|color:%233f3f3f\u00a0\u00bb use_theme_fonts=\u00a0\u00bbyes\u00a0\u00bb][vc_column_text]<strong>B.Nusgens\u00a0&amp; P.Mineur<\/strong><\/p>\n<p>Dans la plupart des cellules, le transfert de signaux issus du milieu ext\u00e9rieur vers les diff\u00e9rents \u00e9l\u00e9ments de la machinerie cellulaire contr\u00f4lant les fonctions vitales telles l&rsquo;adh\u00e9sion, la migration, la synth\u00e8se de produits, la multiplication et la survie utilise l&rsquo;ion calcium comme second messager intracellulaire. Un accroissement transitoire de la concentration de calcium intracellulaire est g\u00e9n\u00e9r\u00e9 par l&rsquo;ouverture de canaux membranaires et\/ou par un relargage r\u00e9versible de cet ion \u00e0 partir des r\u00e9servoirs internes. La plupart des r\u00e9cepteurs membranaires, y compris les r\u00e9cepteurs activ\u00e9s par les signaux m\u00e9caniques, utilisent cette voie sensible de signalisation.
