Boland<\/strong><\/p>\n<p>Quelques \u00e9tudes \u00e9pid\u00e9miologiques ont mis en \u00e9vidence une association entre l&rsquo;exposition \u00e0 des champs \u00e9lectromagn\u00e9tiques EBF (1-100 Hz) et des pathologies neurod\u00e9g\u00e9n\u00e9ratives. En outre, des agressions oxydantes induites par les radicaux libres participent aux dommages tissulaires associ\u00e9s \u00e0 de telles maladies. Une fois qu&rsquo;ils sont produits en exc\u00e8s, les radicaux libres peuvent alt\u00e9rer beaucoup de constituants cellulaires et entra\u00eener la mort cellulaire. C&rsquo;est la raison pour laquelle notre laboratoire a effectu\u00e9 des recherches afin de <strong>d\u00e9terminer si les champs \u00e9lectromagn\u00e9tiques EBF<\/strong> pouvaient avoir des <strong>effets sur des cellules c\u00e9r\u00e9brales en culture<\/strong> en combinaison avec une agression cellulaire induite par des <strong>radicaux libres<\/strong> (stress oxydatif).<\/p>\n<p>Nous avons montr\u00e9 (A. Boland, Bioelectromagnetics, 2002, 23:97-105) que, quand les cellules c\u00e9r\u00e9brales en culture \u00e9taient expos\u00e9es \u00e0 des champs \u00e9lectromagn\u00e9tiques intermittents de forte intensit\u00e9 (2.5 &#8211; 5 mT) en plus d&rsquo;un stress oxydatif induit par de l&rsquo;oxyde nitrique (NO), il se produit une augmentation significative de mort cellulaire, allant de 11 \u00e0 23%. Les causes de cette augmentation restent \u00e0 d\u00e9couvrir.<\/p>\n<p>Quand le NO est produit, il peut r\u00e9agir avec un autre radical libre, l&rsquo;anion superoxyde (O2-\u00b0) , entra\u00eenant la formation de peroxynitrite (ONOO-), un autre composant oxydant capable de tuer les cellules. Par cons\u00e9quent, afin de savoir si les champs \u00e9lectromagn\u00e9tiques sont capables de moduler la cytotoxicit\u00e9 induite par le peroxynitrite, nous avons incub\u00e9 des cellules d&rsquo;hippocampe en culture en pr\u00e9sence de SIN-1, un donneur de ONOO-, en association avec des champs \u00e9lectromagn\u00e9tiques 50 Hz ininterrompus ou intermittents (0 &#8211; 5 mT).<\/p>\n<p>A partir de nos donn\u00e9es, il appara\u00eet que les CEM, seuls, ne sont pas nuisibles pour les neurones. Toutefois, dans des conditions exp\u00e9rimentales particuli\u00e8res (CEM intermittent de haute intensit\u00e9 combin\u00e9 \u00e0 un stress oxydatif), la mortalit\u00e9 cellulaire peut augmenter. Cette toxicit\u00e9 peut varier selon le type et l&rsquo;intensit\u00e9 du CEM utilis\u00e9. Il est toutefois impossible \u00e0 partir des r\u00e9sultats obtenus \u00e0 ce jour sur des cellules nerveuses en culture de conclure \u00e0 une toxicit\u00e9 potentielle des CEM sur l&rsquo;\u00eatre humain, et d&rsquo;autres exp\u00e9rimentations doivent \u00eatre r\u00e9alis\u00e9es afin de comprendre les m\u00e9canismes d&rsquo;action des CEM de fr\u00e9quence extr\u00eamement faible et de d\u00e9terminer les effets <em>in vivo<\/em>.[\/vc_column_text][vc_custom_heading text=\u00a0\u00bb1999-2000&Prime; font_container=\u00a0\u00bbtag:h3|text_align:left|color:%233f3f3f\u00a0\u00bb use_theme_fonts=\u00a0\u00bbyes\u00a0\u00bb][vc_column_text]<strong>V.Seutin &amp; A. Boland<\/strong><\/p>\n<p>Nos pr\u00e9c\u00e9dents travaux ont montr\u00e9 que l&rsquo;utilisation d&rsquo;un champ \u00e9lectromagn\u00e9tique (CEM) intermittent (\u00ab\u00a01 min ON + 1 min OFF, r\u00e9p\u00e9t\u00e9 10 fois ) induit une augmentation de 200 % d&rsquo;un marqueur g\u00e9n\u00e9ral du stress dans les cellules (c-fos). En raison du fait que ce marqueur peut induire diverses r\u00e9ponses cellulaires lorsqu&rsquo;il est activ\u00e9, nous avons tent\u00e9, d&rsquo;une part de d\u00e9terminer l&rsquo;effet d&rsquo;une application r\u00e9p\u00e9t\u00e9e d&rsquo;un tel CEM intermittent (\u00ab\u00a01 min ON + 1 min OFF, r\u00e9p\u00e9t\u00e9 10 fois toutes les 2 heures, 6 fois par jours durant 48 heures) sur des cultures de cellules c\u00e9r\u00e9brales et, d&rsquo;autre part, d&rsquo;\u00e9valuer l&rsquo;effet du m\u00eame CEM en combinaison avec un stress oxydatif induit par un radical libre, le monoxyde d&rsquo;azote (NO).<\/p>\n<p>Les r\u00e9sultats ont montr\u00e9 que :<\/p>\n<p>1. L&rsquo;exposition de cellules c\u00e9r\u00e9brales en culture \u00e0 diff\u00e9rentes intensit\u00e9s (0 \u00e0 5 mT) de CEMs intermittents ne produit aucune modification de la mortalit\u00e9 cellulaire. Il semble d\u00e8s lors que, par eux-m\u00eames, de tels CEMs ne sont pas pr\u00e9judiciables \u00e0 la survie des cellules c\u00e9r\u00e9brales en culture.<\/p>\n<p>2. Les r\u00e9sultats obtenus lorsque les cellules sont expos\u00e9es simultan\u00e9ment aux CEMs intermittents (0 \u00e0 5 mT) et au stress oxydatif induit par le NO ont montr\u00e9 que les intensit\u00e9s les plus faibles (0 &#8211; 0.45 mT) n&rsquo;ont aucune influence sur la mortalit\u00e9 cellulaire. Toutefois, pour des intensit\u00e9s plus importantes (2.5 &#8211; 5 mT), nous observons une augmentation significative de la mort cellulaire variant de 11 % \u00e0 23 %. La cause de cette augmentation reste \u00e0 d\u00e9couvrir.<\/p>\n<p>Il semble d\u00e8s lors que, dans nos conditions exp\u00e9rimentales, les CEMs EBF ne sont pas nocifs vis-\u00e0-vis des cellules in vitro sauf si un stress est d\u00e9j\u00e0 pr\u00e9sent.
