Boland<\/strong><\/p>\n<p>Een aantal epidemiologische studies maakten melding van een verband tussen blootstelling aan ELF EMF&#8217;s (1-100 Hz) en neurodegeneratieve pathologie\u00ebn. Bovendien dragen de oxiderende aantastingen door vrije radicalen bij tot de weefselbeschadigingen die met deze ziekten worden geassocieerd. Zodra er te veel van worden geproduceerd, kunnen de vrije radicalen verschillende bestanddelen van de cellen wijzigen en uiteindelijk tot het afsterven van de cellen leiden. Dat is de reden waarom we in ons laboratorium experimenten hebben <strong>uitgevoerd om na te gaan of ELF EMF&#8217;s een effect<\/strong> zouden kunnen hebben op gekweekte hersencellen wanneer ze met aanvallen van cellen door de <strong>vrije radicalen worden gecombineerd (oxidatieve stress).<\/strong><\/p>\n<p>We hebben aangetoond (A. Boland, Bioelectromagnetics, 2002, 23:97-105) dat het aantal cellen die afsterven aanzienlijk stijgt (van 11 tot 23%) wanneer de gekweekte hersencellen worden blootgesteld aan discontinue EMF&#8217;s (IEMF&#8217;s) met een hoge intensiteit (2,5 &#8211; 5 mT) in combinatie met oxidatieve stress ten gevolge van nitrietoxide (NO). Het is nog niet duidelijk waaraan deze stijging toe te schrijven is.<\/p>\n<p>Wanneer er NO wordt geproduceerd, kan deze stof met een andere vrije radicaal reageren: anion superoxide (O2-\u00b0). Dit leidt tot de vorming van peroxynitriet (ONOO-), een andere oxiderende samenstelling die cellen kan doden. Om te weten te komen of EMF in staat is om cytotoxiciteit die door peroxynitriet wordt veroorzaakt, te moduleren, incubeerden we gekweekte hippocampuscellen in aanwezigheid van SIN-1, een ONOO- donor, in combinatie met ononderbroken of onderbroken elektromagnetische velden van 50 Hz (0 &#8211; 5 mT).<\/p>\n<p>Op basis van onze gegevens blijkt dat elektromagnetische velden alleen niet schadelijk zijn voor de neuronen. In bijzondere experimentele omstandigheden (intermitterende elektromagnetische velden met hoge dichtheden in combinatie met een oxidatieve stress) kan de cellulaire mortaliteit evenwel toenemen. Deze toxiciteit kan vari\u00ebren volgens de aard en de dichtheid van het gebruikte elektromagnetische veld. Het is evenwel onmogelijk op basis van tot dusver verkregen resultaten op neuronen in cultuur conclusies te trekken over een mogelijke toxiciteit van elektromagnetische velden op de mens, en verdere studies zijn noodzakelijk om de mechanismen van de activiteit van elektromagnetische velden met extreem lage frequentie te begrijpen en de effecten in vivo te bepalen.[\/vc_column_text][vc_custom_heading text=&#8221;1999-2000&#8243; font_container=&#8221;tag:h3|text_align:left|color:%233f3f3f&#8221; use_theme_fonts=&#8221;yes&#8221;][vc_column_text]<strong>V.Seutin &amp; A. Boland<\/strong><\/p>\n<p>Onze vorige studies hebben aangetoond dat het gebruik van een intermittent elektromagnetisch veld (EMF) (&#8220;1 min ON + 1 min OFF, 10x herhaald) een 200% stijging veroorzaakt van een algemene stressmarker in de cellen (c-fos). Aangezien deze marker bij activatie diverse celresponsen kan induceren hebben wij getracht om enerzijds het effect te bepalen van een herhaalde applicatie van zulk intermittent EM-veld (&#8220;1 min ON + 1 min OFF, 10x herhaald om de 2 uur, 6 maal per dag gedurende 48 uur) op hersencelculturen, en anderzijds de invloed van hetzelfde EM-veld te evalueren in combinatie met een door een vrije radicaal ge\u00efnduceerde oxidatiestress, stikstofmonoxide (NO).<\/p>\n<p>De resultaten hebben aangetoond dat :<\/p>\n<p>1. Blootstelling van een cultuur van hersencellen aan verschillende intensiteiten (0 \u00e0 5 mT) van intermittente EM-velden geen invloed heeft op de celmortaliteit. Zulke EM-velden hebben dus blijkbaar geen nadelige invloed op hersencellen in cultuur.<\/p>\n<p>2. Bij cellen die tegelijk aan intermittente EM-velden (0 \u00e0 5 mT) en aan een door NO ge\u00efnduceerde oxidatiestress zijn blootgesteld hebben de zwakste intensiteiten (0 &#8211; 0.45 mT) geen enkele invloed op de celmortaliteit. Bij grotere intensiteiten (2,5 &#8211; 5 mT) zien we wel een significante stijging van de celdood gaande van 11% tot 23%. De oorzaak van die stijging is nog onbekend.<\/p>\n<p>Kortom, in onze experimentele voorwaarden zijn de EM-velden LFE niet schadelijk voor cellen in vitro behalve als er reeds een stress aanwezig is.
